Allosteric properties of skeletal muscle pyruvate kinase.

چکیده
The phenylalanine inhibition of skeletal muscle pyruvate kinase has been further studied. Even at high levels of the inhibitor the plots of reaction rates as a function of phosphoenolpyruvate concentration give hyperbolic curves; the inhibition by phenylalanine is of the mixed type. The enzyme exhibits a homotropic cooperative effect with respect to this inhibitor. This property is strongly dependent on PH. Serine, cysteine, and alanine reversed this inhibition. The reactivating amino acids greatly increase the IO.6 but do not alter the cooperativity of the inhibitor. These effects were not found with 2%phosphoglyceric acid, an isosteric inhibitor. In fact, the inhibition is independent of pH and it is not reversed by the amino acids. Furthermore the enzyme follows Michaelis-Menten kinetics with respect to this inhibitor. The reassociation of the enzyme from its subunits markedly decreases the homotropic cooperative effect of phenylalanine. It is concluded that this amino acid is an allosteric inhibitor.

Regulatory Properties of Skeletal-Muscle Pyruvate Kinase
Hitherto it has been generally supposed that pyruvate kinase (EC 2.7.1.40) has no significant role in the regulation in vivo of glycolytic flux in skeletal muscle (Carbonell et al., 1973). This supposition, apart from providing an explanation for the apparent lack of potentially regulatory allosteric interactions by skeletal-muscle (M-type) pyruvate kinase, is in agreement with the idea that gl...
متن کاملL-Phenylalanine inhibition of muscle pyruvate kinase.
The allosteric inhibition of M1-type pyruvate kinase from rabbit skeletal muscle by phenylalanine is reciprocally dependent on Mg2+ and phosphoenolpyruvate concentrations. At pH 8, phenylalanine acts as a competitive inhibitor with respect to Mg2+ and phosphoenolpyruvate, and vice versa. Phenylalanine introduces sigmoidicity into the dependence of the reaction velocity on [Mg2+]. In vitro kinet...
متن کاملThe Effects of Pyruvate Dehydrogenase Kinase 4 (PDK4) Inhibition on Metabolic Flexibility during Endurance Training in Skeletal Muscles of Streptozotocin-induced Diabetic Rats
Background:Metabolic flexibility is the capacity of a system to adjust fuel (primarily glucose and fatty acids) oxidation based on nutrient availability. Pyruvate Dehydrogenase Kinase 4 (PDK4) is one of the main enzymes that play a critical role in metabolic flexibility. In current study, we examined PDK4 inhibition along with exercise training (ET) on the gene expression of Es...
متن کاملAllosteric properties of yeast pyruvate kinase studied by site-directed mutagenesis.
The enzyme from most sources is a tetramer of identical subunits each about 500 residues in length. The enzyme has an absolute requirement for monovalent and divalent cations (usually K+ and Mg2+) which act to coordinate and orientate the substrates prior to catalysis. A number of isoenzymes of pyruvate kinase are found to exist in vertebrate tissues. Their kinetic and regulatory properties ref...
متن کاملFactors affecting the level and activity of pyruvate kinase from Coprinus lagopus sensu Buller.
The specific activity of Coprinus lagopus pyruvate kinase is high under conditions of glycolysis but low under conditions of gluconeogenesis. Only a single form of the enzyme was detectable, the properties of which resemble in a number of respects those of the allosteric forms of pyruvate kinase. These properties include co-operative interactions with phosphoenolpyruvate (PEP), ATP and fructose...
